Spurs took control with second-half goals from Son Heung-min, Jan Vertonghen and Fernando Llorente and Dortmund are 7/1 in the Champions League odds to complete an unlikely comeback.
Keeping a clean sheet at Wembley means one away goal would leave Dortmund needing at least five goals to have any hope of progressing and Spurs have been cut to 14/1 in the Champions League outright betting ahead of the trip to Germany.

Borussia Dortmund vs Tottenham Betting Tips
Here are our best tips for the match for which we also have a range of other markets available:
Tottenham had never won a home knockout tie in the Champions League prior to the first leg and they were arguably second best in the first half at Wembley, where they were without influential duo Harry Kane and Dele Alli.
Son made the breakthrough as he scored his 11th goal in 12 games early in the second half to continue his love affair with playing against Dortmund as that was the ninth time he has scored against them in 11 games.
Vertonghen, who was playing at left wing-back, capped a man-of-the-match performance with a second goal in the 83rd minute and then three minutes later Llorente gave Spurs vital breathing space by nodding home from a corner with his first touch of the game.
Manager Maurico Pochettino hailed his players as "heroes" but predictably warned that the tie is not yet over.
"It's only the first leg, it's only the first half of the tie - 3-0 is a very good result but when you have a team like Borussia Dortmund, always you need to respect and we need to think that we need to go to Dortmund with the idea to win," he said. "We are happy now but still a lot of work to do."
Tottenham's domestic form should ensure there is no complacency, with Spurs having taken just one point from three Premier League matches since the first leg took place.
A 2-1 defeat at Augsburg on Friday will not have helped Dortmund's confidence though and they have won just one of their last seven games in all competitions, drawing four of them.
With Spurs having the luxury of being able to sit back and defend, we think a draw at 27/10 looks the most likely result to kick off our Borussia Dortmund vs Tottenham predictions.
First goalscorer tips
It is rare that Kane is not the favourite to open the scoring but there are two Dortmund players - Marco Reus and Paco Alcacer - ahead of the England striker in the first goalscorer odds at 4/1.
Alcacer had gone eight games without scoring until grabbing a late goal on Friday so we'll rule him out. Reus is in form but we still feel the reliable Kane is a better option at the bigger price of 9/2 given that Dortmund are expected to leave gaps at the back as they push for the three goals they need.
Anytime goalscorer tips
Raphael Guerreiro was initially signed by Dortmund as a left-back but has been playing as a winger and has impressed this season, especially in the Champions League.
He bagged a brace against both Atletico Madrid and Monaco and has been on target twice in the Bundesliga in 2019 so he looks a decent option in the anytime goalscorer odds at 9/2.
Where the game will be won or lost
Dortmund's results this season will give them some hope that they can still turn this tie around despite their recent wobble.
They have scored four goals on six occasions in the Bundesliga - twice as many as any other team - while they also enjoyed a 4-0 win at home to Atletico Madrid during the Champions League group stage.
Dortmund have netted at least two goals in every home game in the league and have had scored three first-half goals twice, so the warning signs are there for Tottenham.
The return of Kane from injury is a major boost though and Spurs will fancy their chances of getting at least one goal which would leave the German side with a mountain to climb.

Dortmund have no choice but to go all-out attack and with Spurs having the quality to pick them off on the break, focusing on the goals markets seems an obvious punting plan.
To complete our Borussia Dortmund vs Tottenham betting tips, we'll take at least four goals in the match at 7/5 with a speculative punt on a 2-2 draw at 12/1.
Tottenham will be without Dele Alli and Kieran Trippier. Alli is in the final stages of his recovery from a hamstring injury while Trippier is being rested, with both men staying in London.
Harry Winks (hip) and Eric Dier (tonsillitis) have travelled and will be assessed ahead of the game.
Dortmund's Christian Pulisic is a doubt with a thigh injury.
Traders' Tips - View from the trading floor
Dortmund looked the better team in the first half at Wembley and although they look to have too much to do to turn the tie around, they should at least win the second leg on home soil.
